Sample Treatment with Trypsin for RT-LAMP COVID-19 Diagnosis

The SARS-CoV-2 coronavirus is responsible for the COVID-19 pandemic resulting in a global health emergency. Given its rapid spread and high number of infected individuals, diagnostic tool rapid, simple, cost-effective detection was essential. In this work, we developed test, that incorporates human internal control, based on Reverse Transcription Loop-Mediated Isothermal Amplification (RT-LAMP). When working with synthetic RNA, optimized RT-LAMP assay has sensitivity 10 viral copies can be detected by fluorescence less than 15 min or naked eye 25 using colorimetric RT-LAMP. To avoid RNA extraction step, pre-treatment sample optimized. Subsequently, validation performed 268 trypsin treated samples (including nasopharyngeal, buccal, nasal exudates) amplified to evaluate specificity comparison RT-qPCR extracted samples. results showed 100% Ct ≤ 30. inexpensive extraction-free procedure may an alternative test could applied adapted detect other viruses present saliva nasopharyngeal higher antibody test.
